There are several locations from which you can take a vacuum signal. Intake manifold, rocker cover, block- you name it. And yes, those pumps work on vacuum pulses- that's why there's 3 ports- vacuum, fuel inlet, fuel outlet. The engine vacuum pulses drive a diaphragm inside the pump. The diaphragm pulses, drawing fuel in thru one 1-way valve, and pushing it out another.

Every engine creates vacuum pulses. These pulses are generated by the piston moving up and down in the cylinder. This results in positive (pressure) and negative (vacuum) pulses.
